(A) U251 and T98G cells were treated with increasing doses of TMZ (0–25 μM) for 48 hours. Cell viability was assessed by Trypan blue exclusion test and expressed as number of cells. (B) MET for 24, 48 and 72 h. Cell viability was assessed as previously described.U251 and T98G cells were treated with 25 μM TMZ and/or 10 mM MET for 24, 48 and 72 h. Cell viability was assessed as previously described. (C) Real time-PCR for pro-apoptotic Bax and Bad genes 48 h after treatment with TMZ and/or MET. Data were normalized for β-actin and ΔΔct expressed as Fold Of Induction (FOI). (D) Real time-PCR for anti-apoptotic Bcl-2 gene 48 h after treatment with TMZ and/or MET. Data were normalized for β-actin and ΔΔct expressed as Fold Of Induction (FOI).
