The selective expansion of red pulp macrophages (F4/80+) and increased expression of PDL1 on all splenic myeloid populations by MIS416 required IFN-γ. (a,b) Splenocytes were isolated from EAE mice on day 22 post immunization and from healthy mice 15 days post MIS416 treatment initiation (24 hours post dose 3), and the total number of each splenic myeloid subset and the expression of PDL-1 (geoMFI) were assessed by flow cytometry (gating strategy in Suppl Fig. 3a).
