Single-cycle RT reaction on a noncoding RNA domain. In these experiments, a fragment from a long noncoding RNA (Domain 3 from lncRNA RepA [Liu et al. 2017]) was used as template, and any dissociated RT was trapped by the addition of a trap duplex (see Materials and Methods) upon initiating reaction. (A) Representative denaturing acrylamide gel of products. The “ctl” lanes indicate conditions in which RT is incubated with a vast excess of trap before being presented to the template.
