a) HA was modified with AD or CD, which self-assemble into shear-thinning and self-healing gels. Likewise, cholesterol on miR-302-chol interacts with CD to sustain release from the gel. b) Rhodamine/CD-HA interactions lead to quenching of rhodamine fluorescence, but the fluorescence is recovered by titration of cholesterol-modified miR-302 into the system and displacement of rhodamine complexes, indicating complexation between cholesterol and CD. c) Release of cholesterol-modified miR-302b and miR-302c (210 µM of each) from gels (5 wt%) in 1.5 mL microcentrifuge tubes over three weeks quantified by RiboGreen, a commercially available RNA quantification kit (mean ± SD, n=3).
