(A) Amniotic cells were seeded in 6-wells plate and incubated with complete IMDM medium supplemented with 10% FBS (C) or hepatic differentiation medium (HD) for at least 30 days. At indicated times, mRNA expression of ALBUMIN, α-FETOPROTEIN (α-FP), α1-ANTITRYPSIN (α1-AT) and CYP7A1 was determined. (B) hAECs were plated in IMDM 10% FBS (C) or treated with hepatic differentiation medium (HD) and lysated for RNA extraction over 20 days of treatment. In (A) and (B) total RNA was extracted as described in Material and Methods and hepatic mRNAs were quantified with real time RT-PCR. CYCLOPHILIN and GAPDH were used as internal standards.
