Ellman’s tests confirmed the successful reduction of analysed PDIs. Upon DTT treatment and buffer exchanges, the percentage of reduced cysteine residues was tested by Ellman’s test. The concentrations of detected free disulfide groups were taken into the relation to the calculated concentration, which was taken from the protein concentration (measured by the absorption at 280 nm) multiplied by the numbers of cysteine residues in the individual PDIs. These concentrations were set to 100% of reduced cysteine residues. PDIA6 was tested in two experimental serials, ones together with PDIA1 and PDIA3, and ones with the indicated TRAP mutants. All tests were performed at least three times independently.
