PHF8 knockout by the CRISPR-Cas9 system in 293T cells reduces hypoxia responses. A. Schematic illustration of the strategy for knocking out PHF8. Control (Ctrl) or PHF8 knockout 293T cells (sgPHF8) were incubated under normoxia (N) or hypoxia (H; 1% O2) for 24 h. The protein and RNA levels of the indicated genes were assayed by western blotting and RT-PCR, respectively. In the control cells, gene expression under hypoxia vs. normoxia was compared; in the knockout cells, gene expression was compared to that in the respective control cells (hypoxia or normoxia). The standard deviation (S.D.) was obtained from at least three independent experiments. The Student’s t-test was used to calculate significance. *: p < 0.05; **: p < 0.01.
