Effect of the prevailing thiamin level (DEF and OS) on euchromatin and heterochromatin markers on −266 to −137 (I) and −131 to +10 (II) regions of Slc25a19 promoter in PAC 266-6. The cells were maintained in thiamin-DEF and -OS media for 10 days, and then, formaldehyde cross-linked chromatin was immunoprecipitated using specific anti-histone H3, anti-H3K4me3, anti-H3K9Ac, and anti-H3K27me3 antibodies, followed by qPCR analysis of the mouse Slc25a19 promoter spanning the region −266 to −137 and −131 to +10 as described in materials and methods. Data (means ± SE) were normalized relative to input DNA and expressed as percentage of enrichment relative to histone (H3) (*P < 0.05 and **P < 0.01).
