U2OS cells stably expressing HA‐tagged Myo1C WT were depleted of single importins (A) or their combinations (B) using commercially available siRNAs (Sigma). After knockdown, cells were fixed and stained with HA antibody (HA‐7, Sigma) to visualize cellular localization of the myosin construct. As a control, a commercially available mix of random siRNAs was used (CTRLsi). Scale bars, 20 μm.

Efficiency of knockdown of importins 5, 7, and β. After siRNA transfection, cells were grown for 4 days, after which total RNA was purified and converted into DNA by means of reverse transcription, and levels of specific mRNAs were determined by qPCR analysis using GAPDH as a housekeeping reference gene. Data are mean ± SD from three biological replicates.
