Binding titration of T1 TCR or SCLs with monomeric or tetrameric MART-1·HLA-A2 ligand. T1 or T1-SCLs were expressed on yeast surface and stained with varying concentrations of monomeric MART-1·HLA-A2 (biotinylated), followed by streptavidin-PE (A) or streptavidin-PE tetramers of biotinylated MART-1·HLA-A2 (B). Resultant mean fluorescence units (MFU) were plotted against ligand concentration to obtain binding curves.
