Table 3.

Evaluation of extraction methods and sample mass on heroin samples of varying purities.

Sample Sample mass (g) Extraction method Cycle threshold (Cp)
WP-1 1.0 Maxwell 32.09
QP 32.31
QS 30.89
2.0 Maxwell 34.38
QP 33.72
QS 32.22
BPf-1 1.0 QP 33.10
QS 29.47
2.0 QP 34.2
QS 30.49
BPc-1 1.0 QP 27.64
QS 29.67
2.0 QP 28.77
QS 30.77
NTC >35.00
>35.00

Lower crossing point (Cp) values indicate higher DNA concentrations. The no-template controls (NTC) are also included and exhibit Cp values > than 35, which indicates an unreliable signal to accurately determine the presence of significant amplification. Note, the Maxwell RNA LEV method was only used to extract DNA from sample WP-1 because the remaining samples lacked the necessary mass to perform the three extraction methods.
