Effect of IFN-λ expression on antiviral activity of exogenous IFN-α by immunostaining for HCV core. HCV-infected Huh-7.5 cells (C/T) after 6 days were transfected with mammalian expression plasmid that contained full-length cDNA clone for IFN-λ1 (250 ng/mL), IFN-λ3 (250 ng/mL), IFN-λ4 (50 ng/mL), or pcDNA3.1 (250 ng/mL) vector alone for 24 hours and then treated with 100 IU/mL IFN-α for another 24 hours. HCV core protein expression (brown) was detected by immunohistochemistry. Percentage of HCV core positive cells were counted in five different high-power fields (40×).
