ABT-199 interacts synergistically with FP to induce apoptosis in unfavourable risk multiple myeloma cells. (A) NCI-H929 cells were incubated with ABT-199±FP for 24 h, after which cell death was analysed by flow cytometry after staining with 7-AAD. **P<0.01; (B) H929 and OPM-2 cells were exposed (24 h) to varying concentrations of FP±ABT-199 at a fixed ratio (1 : 5), after which the percentage of Annexin V+ cells was determined. Combination Index (CI) values less than 1.0 denote a synergistic interaction. The results are representative of three separate experiments; (C) H929 cells were treated with 500 nM ABT-199±100 nM FP for 24 h, after which slides were prepared and stained with antibodies against activated caspase-3, followed by Alexa Fluor 594-conjugated secondary antibody with DAPI counterstaining (left panels, × 200 magnification).
