The FP/ABT-199 regimen is highly active against unfavourable prognosis primary MM cells. Isolated primary CD138+ cells from a patient with either no (A, left panel) or favourable (for example, t(11;14)) (A, right panel) FISH abnormalities were exposed to 200 nM ABT-199±75 nM FP for 24 h, after which slides were prepared and stained with antibody against activated caspase-3, followed by Alexa Fluor 488-conjugated secondary antibody with DAPI counterstaining (× 200 magnification). Parallel studies were performed on three samples obtained from patients with unfavourable karyotypes (del 17p; B, left and central panels) or PCL (B, right panel).
