Figure 3. — Linearity of qPCR reactions utilizing genomic DNA, NUversa or plasmid pNUversa. Genomic DNA (blue), NUversa (orange) or pNUversa (green), were serially diluted to obtain seven (genomic DNA and NUversa) or eight (pNUversa) standards (detailed in Methods) spanning 2 through 4.29 × 10⁵, 5 through 1.13 × 10⁶ and 4 through 8.52 × 10⁶ genome equivalents, respectively. Genome equivalent standards were utilized as template in serotype-specific qPCR reactions targeting (A) serotype 1, (B) serotype 3, (C) serotype 4 or (D) serotype 5. Plots represent the mean of cycles of threshold values obtained from duplicate reactions. Regression equations, coefficient of determination (R²) and reaction efficiency are shown in the insets.
