Proposed model for the function of PGRMC1 in hPSCs. PGRMC1 is expressed on the cell surface and inside the cell. PGRMC1 directly suppresses CycD1, p53 and β-catenin and indirectly suppresses them through suppression of Wnt/β-catenin signaling. PGRMC1 also suppresses β-catenin through activation of GSK-3β or inhibition of Wnt3a expression. PGRMC1-mediated suppression of p53, cyclin D1, and β-catenin inhibits differentiation of hPSCs into multi-lineage cells during early differentiation of hPSCs.
