Absence of TLR2 in DCs impairs Th1 polarization in a DC and CD4+ T cell coculture system. Immature BMDCs from WT and TLR2−/− mice were treated with 5 μg/ml of DnaJ-ΔA146Ply or 100 ng/ml of LPS for 24 h and then cocultured with purified naive CD4+ T cells sorted from spleens of WT and TLR2−/− mice, respectively. (A to C) After 72 h, IFN-γ, IL-17A, and IL-4 levels in culture medium were assayed by ELISA.
