The rimmed clone is a three-dimensional structure. A rimmed clone was generated from tissue stem cells (TSC) that harbored the K5-rTA and TRE-Histone 2B transgenes. The clone was cultured in doxycycline-free medium to identify label-retaining (GFP+) cells. Confocal microscopy was used to generate a three-dimensional rendering of the rimmed clone. The central cobblestone domain is to the left and the rim domain is to the right. Label-retaining cells have green nuclei. Label-diluting cells are GFP negative and are stained with DAPI (blue).
