Figure 3. Administration of purified TSP-1 reduced lung bacterial burden, airspace neutrophil recruitment, lung MPO, and lung NE activity in Thbs1–/– mice when compared with Thbs1–/– vehicle-treated mice.

Figure 3

WT and Thbs1–/– mice (KO) were inoculated with Pseudomonas aeruginosa strain PA14 intratracheally at 106 inoculum and subsequently administered either vehicle or purified TSP-1 (pTSP-1) by i.p. injection. Outcome measurements were obtained 20 hours after infection. (A) Lung CFU/ml, (B) total bronchoalveolar lavage (BAL) cells counts/ml, (C) total BAL PMN counts/ml, (D) lung myeloperoxidase (MPO) activity, and (E) lung neutrophil elastase (NE) activity were measured in WT and Thbs1–/– mice. Each data point represents an individual mouse, n = 7 or 8 mice per group. Lines indicate the median. *P < 0.05, **P < 0.01 by Kruskal-Wallis test with Dunn’s multiple comparisons.
