Cnb1-deficient CD4+ T cells produce more interferon (IFN)γ. (A) Proportion of CD4+ T cells producing IFNγ, IL-2, IL-17, or IL-4 in colonic-lamina propria (LP) isolated from Cnb1fl/fl and Cnb1CD4 mice. (B) Purified naïve CD62LhighCD44neg CD4+ T cells from the spleens of Cnb1fl/fl and Cnb1CD4 mice were cultured under conditions to promote Th0, Th1, Th17, Th2, and Treg cells. T cells were collected after 5 days and stimulated with PMA/ionomycin for 5 h, with the addition of Brefeldin A for the final 4 h. Cells were stained for intracellular IFNγ, IL-17, IL-4, and FoxP3, and the proportion of CD4+ T cells producing these cytokines was assessed by flow cytometry. Representative flow-cytometric dot plots are shown.
