Binding of gPr80 to mA3 and A3G in the cytosol. (A) Virus-expressed and plasmid-expressed gPr80 and Pr65 Gag proteins display similar profiles in transfected 293T cells. gPr80 and Pr80[3Q] were expressed from a CMV promoter. The gPr80 initiation codon was changed from CTG to ATG to improve expression. The Pr80[3Q] coding sequence was modified to replace the three glycosylated asparagines (N), at positions 113, 480, and 505, with glutamines (Q) (33). (B) Coimmunoprecipitation of FLAG-mA3 and FLAG-A3G with gPr80 and Pr80[3Q] in the soluble fraction of transfected 293T cell lysates. Protein interactions were revealed by Western blotting using anti-FLAG monoclonal and anti-p30 polyclonal antibodies.
