Trehalose does not interfere with cellular glucose uptake under various glucose uptake assay conditions. We compared our assay conditions for glucose uptake (A) with those used by DeBosch et al. (B). Non-serum-starved HFFs were untreated (−) or incubated with 100 mM trehalose (Tre) 15 min (A) or 30 min (B) prior to the addition of radiolabeled [1,2-3H]2-deoxy-d-glucose at the indicated levels of total 2DG. Uptake was allowed for 5 min (A) or 6 min (B) before stopping with ice-cold washes. Lysis was completed by incubation with the indicated solutions. The level of radioactivity in cells was assayed by scintillation counting of lysates. In each experiment, the assay was performed on triplicate wells. Error bars indicate standard errors of the means for triplicate wells.
