Lipid headgroups modestly affect the kinetics of Bam complex–mediated OmpA assembly. Urea-denatured OmpA was incubated with SurA and proteoliposomes that contained the Bam complex and the indicated lipids at 30 °C, and the folding of the protein after 0–40 min was monitored as described in the legend to Fig. 4. The effect of varying the thickness/fluidity of the bilayer (A) or the lipid headgroup (B) was examined. The data were normalized to the maximum fraction of the protein that folded during each assembly reaction, which was defined as 1.0. Average values from multiple experiments (symbols) and S.D. values (error bars) at each time point are shown. The rate constant (k) and time required to reach 50% maximal assembly (t½) for each reaction were calculated from single-exponential fits. To facilitate comparisons, the kinetics of assembly of OmpA into proteoliposomes that contained POPC are shown in both A and B.
