TFL1 and TFL2 silencing affects SL RNA synthesis. Newly synthesized RNA was labeled with [³²P-α]UTP in permeabilized procyclic trypanosomes, which were uninduced or gene-silenced for 2 or 3 days, extracted, separated on a 50% urea-6% polyacrylamide gel, and visualized by autoradiography. Numbers on the bottom refer to relative SL RNA signal strengths normalized by the corresponding tRNA signals as determined by densitometry. The numbers represent the average of two independent experiments. On the left, pBR322-MspI marker sizes are indicated.
