Adenovirus-mediated effective transduction of BMP2, Sox9, Smad7 and GFP into the micro-mass formed with C3H10T1/2 cells. (A) C3H10T1/2 cells were seeded as micromasses and infected with Ad-BMP2, Ad-Sox9, Ad-BMP2+Ad-Sox9, Ad-Smad7 and Ad-GFP. Representative bright field and GFP fluorescence fields were recorded at 24 h after infection. Representative results are shown. (B) Western blot analysis of adenovirus-mediated transgene expression. Total cell lysate was collected form the micromass culture at 48 h after infection and subjected to SDS-PAGE. The transgene expression of BMP2, Sox9 and Smad7 was probed with respective antibodies. Ad-GFP infected cells were used as negative controls. GAPDH expression was used as loading controls. Representative results are shown.
