Analysis of EMT features in a panel of breast cancer cell lines and non-transformed human mammary cells. (a) Morphology of epithelial and mesenchymal cells assessed under phase contrast light microscopy. Epithelial cells are characterised by cobblestone-like morphology whereas mesenchymal cells display spindle-like, elongated phenotype. (b) Western blot analysis of EMT markers. Decreased expression of epithelial phenotype marker E-cadherin and enhanced expression of mesenchymal markers N-cadherin, fibronectin and vimentin were found in parallel to morphological features associated with epithelial and mesenchymal phenotypes. GAPDH was used as a loading control. (c) The mRNA levels of the EMT transcription factor SLUG were quantified using SYBR green-based qPCR in the indicated cell lines. MCF-10A (non-malignant), MCF-7, ZR-75-1 and T47-D (non-invasive ERα-positive), MDA-MB-231 and Hs578T (highly invasive ERα-negative) cells.
