Hyperglycaemia-induced, matrix-specific EMT triggers a metabolic switch towards the Warburg effect. (a) Glucose uptake was assessed using the non-metabolisable glucose analogue, tritiated 2-deoxyglucose, in MCF-7 cells with or without FASN silenced with siRNA. The mRNA levels of the indicated metabolic enzymes were detected using SYBR green-based QPCR as shown in b, d, e, g and h. Lactate and citrate levels were measured in cell supernatants using colorimetric based assays (c and f). In all cases, results shown are representative of three independent experiments, each performed in triplicate and are expressed as means of ±SEM.
