Deficiency of H3K4me3 histone methylation modification impedes the activation of hypoxia‐induced LX‐2 cells. LX2 cells were pretreated with 1 mm MTA for 16 h and stimulated with 100 μm CoCl₂. Cells were collected at indicated time. (A) Activation of molecular marker of HSC, α‐SMA, was detected by western blot, and densitometric analysis was performed using pooled data from three such experiments. Results were expressed as mean ± SD, *P < 0.05, **P < 0.01. (B) Increase in vimentin (red) and re‐organization of F‐actin (red, CY3‐labeled phalloidin) were detected with immunofluorescence staining (blue: DAPI, 400×).
