Table 3.

Quantity and quality of the extracted RNA.

Cell fraction Absolute RNA amount (ng) RNA amount per Cell (pg) RIN
Total pancreas 50000 ± 7400 16.6 ± 2.4 4.8 ± 1.1
Acinar 3000 ± 200 16 ± 1.3 8.6 ± 0.17**
Acinar (8 h Caerulein) 2750 ± 396 11.5 ± 0.99 8.1 ± 0.1*
Acinar (72 h Caerulein) 886 ± 112 8 ± 1 8.4 ± 0.09**
Ductal 380 ± 20 12.6 ± 0.8 7.4 ± 0.46*

For RNA from the whole pancreas, the extraction was performed on a small volume of the entire digestion solution (~ 3,500,000 cells), before FACS sorting. For the acinar fraction treated or not with caerulein, the extraction was performed on an average number of 150,000 sorted cells. For the ductal fraction, the number was reduced to 25,000 sorted cells per pancreas. Results are presented as mean ± SEM (total pancreas: n = 3; acinar fraction: n = 7; caerulein-treated acinar fraction 8 h: n = 3; caerulein-treated acinar fraction 72 h: n = 11; ductal fraction: n = 3). Statistical significance was measured using a t-student test and p-values were considered statistically significant when P < 0.05 (

*

P < 0.05,

**

P < 0.01, compared to total pancreas fraction).
