Differentiation of hiPSCs into mesencephalic DA neurons. A, The expression of β3-tubulin (red), tyrosine hydroxylase (TH, green) and Lmx1A (purple) in cultures of dopamine neurons differentiated for 21–27 days was assessed by immunocytochemistry, all cultures were counterstained with a nuclear stain, Hoechst (light blue) (shown here is cell line CA11, Scale bar = 50 µm). B, β3-tubulin-, Lmx1A- and TH-positive cells were quantified by high content imaging. Quantification for β3-tubulin and TH expression was from 18, and Lmx1A from 4 individual experiments including cell lines derived from 4 different control subjects. (shown are means ± SEM).
