(A) GLUT4 translocation to the plasma membrane. mCherry-GLUT4-myc-transfected L6 cells were starved of serum for 4 hr and then incubated in the absence or presence of A77 1726 (200 μM) without or with 2X the amino acid concentrations. Cells were left unstimulated or stimulated with 20 nM insulin for 45 min. After fixation in methanol for 10 min, mCherry-tagged GLUT4 fluorescence was visualized under a Leica LP8 confocal microscope. Arrows denote the mCherry-tagged GLUT4 translocation to the cell membrane. (B) Quantification of the GLUT4 translocation to the plasma membrane. The data represent the mean ± SD from one of three experiments with similar results. *p<0.05; **p<0.01
