Effect of LUF6000 or LUF6096 on dissociation of [125I]I-AB-MECA binding to HEK 293 cell membranes expressing human (a), dog (b), rabbit (c), or mouse (d) A3ARs. Membranes (50 μg) were incubated with [125I]I-AB-MECA (~ 0.3 nM) for 2 h at room temperature after which dissociation was initiated by adding 100 μM NECA mixed with either vehicle or modulator compound. The fraction of specific binding at various times after the addition of NECA is shown. The data were fitted to a two-phase exponential decay model. Data are the mean ± SEM of five to eight experiments performed in triplicate. See Table 1 that reports dissociation rate constants
