Identification of an ING3 transcriptional network. (A) The expression of ING3 was silenced in the LNCaP cell line using two different siRNA in four independent biological replicates, total RNA isolated, and hybridised on Illumina Human HT-12 arrays. The top 25 downregulated and top 25 upregulated genes were plotted. (B) Silencing of ING3 was assessed by immunoblotting using the indicated antibodies. (C) Silencing of ING3 was confirmed by real-time qPCR. (D–J) Hits from the microarray analysis were validated by standard real-time qPCR on reverse transcribed total RNA isolated from control (Scramble siRNA) or ING3-silenced (siING3 no.1 and siING3 no.2) LNCaP cells. Expressed values are the average of three biological replicates, each performed in technical triplicate, normalised to the expression of HPRT1, with s.e.m. as error bars (*P<0.05, **P<0.01, ***P<0.001, and NS indicates not significant, as calculated by t-test).
