Early intramuscular transgene expression kinetics of 5 different adeno-associated virus (AAV) capsids. Albino C57BL/6 mice (n = 4 per group) were injected intramuscularly with 1 × 1011 vector genomes of an AAV vector expressing firefly luciferase (Luc) packaged with either AAV6, AAV6.2FF, AAV8, AAV9, or AAV-DJ capsid. A, In vivo luciferase images were obtained 0, 1, 3, 7, and 14 days after AAV administration to demonstrate vector distribution. B and C, Relative photon emission produced by luciferase from the muscle exclusively (quantified from the lateral view; B) or the muscle plus the liver (quantification of the ventral view; C) of each serotype of AAV was quantified at various time points from 1 to 112 days after injection.
