CotE binds to mucin via GlcNAc and GalNAc. The binding of proteins corresponding to the N-terminal (rCotEN) and C-terminal (rCotEC) domains of CotE to mucin or ligands was measured by enzyme-linked immunosorbent assay (ELISA). Mucin or ligands were first coated onto ELISA plates and either rCotEN or rCotEC then applied. Bound protein, after washing, was measured by ELISA. The experiment was independently repeated 3 times.
