Memory and naïve T cells express similar levels of vitamin D response machinery. RNA was purified from naive and memory CD4+ T cells from the peripheral blood of healthy controls before and after stimulation with antiCD3/CD28 beads. Regulators of response to 1,25(OH)2D3 including (A) Vitamin D receptor (VDR), (B) Retinoid X Receptor (RXR), (C) VDR co-regulators: DRIP205, NcoA1, NcoR1 and NcoR2 were measured by qPCR. mRNA expression for each gene is shown relative to matched memory T cells. VDR protein in T cells was also measured by flow cytometry before and after stimulation with PMA and ionomycin. Stimulated T cells were defined as CD69+. Statistical significance was tested by repeated measures two-way ANOVA. * = P < 0.05, ** = P < 0.01.
