Depleting sirtuin 4 (SIRT4) sustains glycolytic activity during physiological resolution of acute inflammatory response. Control knockdown and SIRT4 knockdown THP-1 cells were stimulated with 1 µg/ml lipopolysaccharide (LPS) for indicated times. (A) Cellular glycolytic activity was assessed by the conversion of d-[5-³H(N)]glucose to ³H₂O, the changes of ³H₂O generation rate in control and SIRT4 knockdown cells were plotted. (B) Changes of lactate dehydrogenase (LDH) activity and (C) Changes of ratio of cellular lactate to pyruvate level in both control and SIRT4 knockdown cells were assayed using colorimetric assay kit.
