Kyte and Doolittle plots representing hydrophobicity change as a consequence of amino acid substitution/s in the TROL presequence [23]. Amino acids 67–78 of the partially conserved N-terminal part of the presequence around the predicted transit peptide cleavage site, AKSLTYEEALQQ, were substituted as follows: e1: 67Ala→67Ile, e2: 71Thr→71Asn, e3: 72Tyr→72Val, e4:73Glu74Glu→73Gln74Gln, e5: 76Leu→76Thr, e6:78Gln→78Val. For the e6 substitution, a polarity check according to Zimmerman was performed [24].
