Table 2.

Measured transcellular CDF flux across Caco-2 monolayers, and CDF amounts in donor compartment and cells at 120 minutes following initial exposure to 5 μM CDFDA (Control), and with either 100 μM MK-571, glyceollin or genistein.

Flux was calculated from linear regression of the amount of CDF transported versus time from 30 to 120 minutes (zero-order and steady-state kinetic assumption). SEM refers to standard error of the mean. For each transport direction, there were 48 filters in control experiments and 9 filters for each agent. For a given transport direction, flux and cell content were significantly different from control in the presence of all three agents (p < 0.05) according to a 1-way ANOVA followed by Tukey’s test for multiple comparisons. For a given transport direction, donor amounts in the presence of either MK-571 or glyceollin were not significantly different from control; whereas, donor amounts were different for genistein.
