The primary analyses conducted in ALSPAC/ARIES. In stage 1, 118 associations between published type 2 diabetes SNPs and genome-wide methylation were identified. In stage 2, a further 226 mQTLs were identified between 202 additional independent non–type 2 diabetes SNPs and the CpGs identified in stage 1. DIAGRAM data were then used to assess the relationship of these independent mQTLs with type 2 diabetes disease risk in order to strengthen causal inference that DNA methylation is acting as a mediating mechanism.
