Normal γ-tubulin localization is not perturbed in the conditional tubg1 mutants. Exponentially growing cells at 30°C were either shifted at 18°C for 8 h, fixed in methanol, and then processed for immunolocalization with the use of the mouse monoclonal GTU-88 followed by Cy3-conjugated goat anti-mouse antibodies. DNA was stained with 10 μg/ml Hoechst for 1 h. Arrows indicate dividing cells.
