Flow cytometric analysis of DNA content of the cold sensitive tubg1 mutants. Wild-type cells (A), gtb1Δ cells expressing TUBG1 (B), and wild-type cells expressing TUBG1, and the dominant (D–I) and the recessive (J–O) cold-sensitive cells were grown to mid-log phase at 30°C and then shifted to 18°C for 10 h. Cells were fixed with ethanol and stained with the fluorescent DNA dye SYTOX Green. For each strain, 10,000 events were analyzed. The relative fluorescence corresponding to 1N, 2N, 4N, and 8N is indicated on the x-axis. The profiles reveal a striking difference between wild-type controls and tubg1 mutants.
