Wild type, full-length Spn1 can bind DNA and nucleosomes. (A) DNA (2.5 μM 601–147) or (B) nucleosomes assembled with histone octamer on 601–147 DNA were incubated with a 2- or 4-fold molar excess of Spn1 (WT) or Spn1141–305 as indicated, for 30 min at room temperature. Free DNA and protein complexes were separated by electrophoresis through a native 5% polyacrylamide gel and the gel stained with ethidium bromide. The length in base pairs of select DNA fragments in the electrophoresis markers (M) is shown to the left of the panels. Lanes with DNA (D) or nucleosomes (N) alone are indicated.
