Dynamic modulation of intercellular tether formation and their cytoskeletal organization. (A) 4T1 cells were cultured for 24 hrs on tissue culture dishes coated with 10 µg/ml fibronectin (FN). Cell movement was then monitored by live-cell phase contrast microscopy. Using these movies, we measured the formation, elongation, and shortening of individual tethers, over time. Each curve represents changes displayed by a single tether. The blue curve, and the associated numbers, correspond to the temporal snapshots shown in (A’). Arrowheads denote the measured tether, the features of which are described in Results. Scale bar: 50 µm. (BD) 4T1 cells were cultured for 24 hrs in tissue culture dishes, then fixed and immunolabeled for Tubulin (B), Actin (C), or Vimentin (D). Scale bar: 10 μm.
