Effects of rhSCL on bone mineralization. Bovine bone cores were perfused with calcein and processed for sectioning as described in materials and methods. A: a cross section of a whole bone core (dimensions 10 mm × 5 mm) cultured with daily mechanical loading. B: an example of a bone core loaded in the presence of rhSCL. C: labeling at a higher magnification (20× objective) showing an example of calcein labeling at the trabecular surface (arrowhead) and a labeled osteocyte lacuna (arrow). D: quantification of total calcein labeling; data shown are means of fluorescence intensity ± SE. n = 5 bone cores group, all obtained from the same bovine sternum. Similar results were obtained for 2 independent experiments.
