DC activation does not promote March-I v2 mRNA degradation. DCs were pretreated (or not) with LPS for 1 h, washed, and then incubated in complete medium containing actinomycin D (5 μg/ml) for the indicated time. Cells were harvested, mRNA was isolated, and March-I v2 expression was determined by RT-PCR. The data from each condition were normalized to the expression of GAPDH at each time point. A, March-I v2 mRNA expression in each condition was plotted as a function of time incubated in actinomycin. B, March-I v2 mRNA expression in either untreated or LPS-treated DCs at each time point was normalized to March-I v2 expression at t = 0 (time of addition of actinomycin D).
