Figure 4. — EMT inhibition induced by REC8 overexpression is reversed by REC8 ablation or EGR1 overexpression. REC8-overexpressed BGC823 cells were transiently transfected with REC8 shRNA or EGR1 overexpression plasmid. (A) The mRNA expression levels of EMT markers (SOX10, E-cadherin and vimentin) were assessed using qRT-PCR. (B) The protein levels of REC8, EGR1 and EMT markers (SOX10, E-cadherin and vimentin) were assessed using western blotting. (C) The location of β-catenin in gastric cancer cells was assessed using immunofluorescence assay (magnification, ×400). All the results were expressed as the mean ± SD of three independent experiments with triplicate wells.
