Targeting PDE3A through protein–protein interactions. Conventional PDE3 inhibition activates cAMP-mediated signalling in multiple intracellular compartments, leading possibly to a combination of beneficial and adverse effects. Blocking the protein–protein interactions through which PDE3A is integrated into the SERCA2 complex may stimulate intracellular Ca2+ cycling without the adverse effects.
