Evaluation of LGM2605 inhibition of proinflammatory cytokine release and inflammasome activation in elicited, peritoneal macrophages exposed to asbestos. Elicited macrophages from WT and Nrf2−/− mice were exposed to LGM2605 (50 µM) 4 h prior to asbestos challenge (20 µg/cm2) and cells were harvested and cell culture medium collected at 8 h and 24 h post asbestos exposure. Inflammasome-relevant cytokine release, IL-1β (A) and Il-18 (B); cellular mRNA levels of IL-1β (C) and NLRP3 (D); and caspase-1 activity (E) were evaluated at 8 and 24 h post exposure to asbestos. Samples were run undiluted in triplicate and data are presented as mean ± SEM. Solid lines (―) indicate statistically significant differences between asbestos-only exposure and asbestos plus LGM2605-treatment within each respective cell type and time point.
