Analysis of in vitro DNA damage by (–)-lomaiviticin A (4). (A) (–)-Lomaiviticin A (4) induces DNA DSBs in vitro. (B) Freifelder–Trumbo analysis of DNA damage by (–)-lomaiviticin A (4). Over a 70-fold concentration range, the ratio of SSBs to DSBs induced by (–)-lomaiviticin A (4) was invariant (5.3 ± 0.6):1, indicating that DNA cleavage occurs through a single binding event (“first-order mechanism”).
